CRISPR(Clusteredregularlyinterspacedshortpalindromicrepeats)/CRISPR-associated(Cas)systemsprovidebacteriaandarchaeawithadaptiveimmunityagainstvirusesandplasmidsbyusingCRISPRRNAs(crRNAs)toguidethesilencingofinvADI ngnucleicacids.TheCRISPRsystemconsistsofashortnon-codingguideRNA(sgRNA)madeupofatargetcomplementaryCRISPRRNA(crRNA)andanauxiliarytransactivatingcrRNA(tracrRNA).ThesgRNAguidestheCas9endonucleasetoaspecificgenomiclocusviabasepairingbetweenthecrRNAsequenceandthetargetsequence,andcleavestheDNAtocreateadouble-strandbreak.Thelocationofthebreakiswithinthetargetsequence3basesfromtheNGGPAM(ProtospacerAdjacentMotif).ThePAMsequence,NGG,mustfollowthetargetedregionontheoppositestrandoftheDNAwithrespecttotheregioncomplementarysgRNAsequence.
